Abstract Large amount of waste frying oil are generated throughout the world, particularly in highly populated countries. Discharge of these lipids rich wastes into the sewerage system increase the organic load of the water sources and hamper the wastewater treatment plant. These organic rich wastes can be used as low cost feedstock for microbial conversion into added-value products. In this study, a new microbial strain Bacillus licheniformis NJ1 was isolated from frying oil discharge site and screened for production of lipopetide biosurfactant using frying oil and co-substrate glucose. Microbial strain was identified by 16S rDNA sequence analysis and biosurfactant was characterized by FTIR. Cell growth 0.62 g/L and biosurfactant production -(E24 34%) was achieved using frying oil in media. Glucose 12 g/L, frying oil 19 g/L and sodium nitrate 1.1 g/L was optimized in media by Response Surface Methodology for the highest biosurfactant production -E24 65%. Co-substrate improved biosurfactant production due to higher cell density, quorum sensing and frying oil utilization. Frying oil consumption increased from 33.5 to 72% (2.1 times) in the optimized mixed substrate fermentation. The bioprocess described here could be applied to manufacture large scale added-value products (biosurfactant, enzymes, PHAs, lipids etc.) economically using waste frying oil with low cost sugars derived from agro-processing waste/sugar containing wastes (molasses, canneries waste etc.) from food processing industries fulfilling the requirement of circular economy, where no wastes are generated.
